Microgel encapsulated nanoparticles for glucose-responsive insulin delivery

چکیده

An insulin delivery system that self-regulates blood glucose levels has the potential to limit hypoglycemic events and improve glycemic control. Glucose-responsive systems have been developed by coupling oxidase with a stimuli-responsive biomaterial. However, challenge of achieving desirable release kinetics (i.e., within minutes after elevation duration on order weeks) still remains. Here, we develop glucose-responsive using encapsulated glucose-responsive, acetalated-dextran nanoparticles in porous alginate microgels. The respond rapidly changes concentrations while microgels provide them protection stability, allowing for extended release. This reduces sugar diabetic mouse model at rate similar naked responds 3 days administration similarly healthy animal. With 2 doses containing 60 IU/kg each, are able achieve control mice 22 days.

Glucose-responsive insulin delivery systems.

pH Responsive Polymeric Nanoparticles for Oral Insulin Delivery

Gelatin-eudragit L100 nanoparticles of wet size range 170-563nm were prepared by two step dissolvation method and the effect of different concentrations of eudragit L100 and emulsifying agent sodium lauryl sulphate (SLS) on the particle size were studied. Synthesized nanoparticles were characterized by attenuated total reflectance-fourier transform infrared spectroscopy (ATRFTIR) and the mean s...

Glucose- and temperature-sensitive nanoparticles for insulin delivery

Glucose- and temperature-sensitive polymers of a phenylboronic acid derivative and diethylene glycol dimethacrylate (poly(3-acrylamidophenyl boronic acid-b-diethylene glycol methyl ether methacrylate); p(AAPBA-b-DEGMA)) were prepared by reversible addition-fragmentation chain transfer polymerization. Successful polymerization was evidenced by 1H nuclear magnetic resonance and infrared spectrosc...

Peptide Encapsulated Nanoparticles for Brain Drug Delivery

BBB is acting as a highly selective membrane that separates the circulating blood from brain fluid [1]. This barrier is formed by endothelial cells which are connected to each other at “tight” junctions that create a complete seal between them. Consequently, only lipophilic molecules are able to cross the barrier. Hydrophilic substances, in contrast, are unable to cross the lipid walls unless a...

Non Inflammatory Boronate Based Glucose-Responsive Insulin Delivery Systems

Boronic acids, known to bind diols, were screened to identify non-inflammatory cross-linkers for the preparation of glucose sensitive and insulin releasing agglomerates of liposomes (Agglomerated Vesicle Technology-AVT). This was done in order to select a suitable replacement for the previously used cross-linker, ConcanavalinA (ConA), a lectin known to have both toxic and inflammatory effects i...
